（一）灵敏度高
虽然酶活性调节Elisa试剂盒方法的灵敏度目前并不十分理想，但在酶活性放大Elisa中，检测的灵敏度远比RIA高。根据质量作用定律。即免疫反应所形成的免疫复合物量与反应物浓度成正比。推测所检测的待测物分子数为1。已知1个摩尔浓度含6.02×1023个分子，那么理论推测酶活性放大Elisa方法的zui低检测限可达1.7×10-24mol/L。虽然在实际应用中由于反应条件和试剂纯度以及仪器精度等因素的影响，往往达不到这个水平（大于104个分子），但表明Elisa在灵敏度方面的改进潜力是很大的。
（二）特异性强
从免疫反应的角度来说，Elisa试剂盒与RIA的特异性应该是一致的。但在Elisa方法中，由于作用检测指示剂的酶与其底物的反应有专一性，从而增加了该方法的特异性。
 （三）对仪器设备要求不高，测定成本低
Elisa测定在普通实验室便可进行，常用的仪器设备包括加样器、培养箱、酶标读数器等。按现有价格折算，酶标仪的价格只及液闪仪的1/6至1/4，因此每测定一个样本所需成本不及PIA的1/10至1/16。某些定性Elisa试剂盒方法，还可在野外进行，操作十分方便。
 （四）方法快速、简便
均质酶免疫测定方法操作时，所有反应试剂均在同一体系内进行，不需任何分离步骤，操作十分简便、快速，数分钟便能得出结果。某些定性Elisa试剂盒，如国外的某些奶牛发情鉴定和妊娠诊断Elisa试剂盒，数分钟时间便能完成一次测定。
 （五）试剂保存时间较长
作为检测指示剂用的酶和酶标记物，在低温或干燥条件下相对稳定，可保存半年至数年之久。
 （六）自动化程度高
Elisa由于不存在放射性同位素污染，因此，Elisa试剂盒除加待测样本需要人工操作外，其他各步骤均可用仪器自动化完成。在这种自动化条件下，平均每人每天可完成2000余个样本的检测工作。
